SENSITIZED LANTHANIDE FLUORESCENCE DETECTION OF STEROIDAL HORMONES
2000
Several steroidal hormones with an α,β-unsaturated carbonyl group sensitized the fluorescence of terbium(III) ions: medroxyprogesterone, medroxyprogesterone acetate, methyltestosterone, nandrolone, nandrolone decanoate, progesterone, testosterone, testosterone enanthate, and testosterone propionate, but other steroids which have this chromophore did not give any signal: clostebol acetate, dehydrotestosterone, dydrogesterone, methandienone, and methenolone enanthate. Sensitized fluorescence was used to detect the steroids when eluted with micellar mobile phases of sodium dodecyl sulphate (SDS)-acetonitrile and SDS-pentanol containing 0.01 M Tb(III), with 245 nm and 545 nm as excitation and e…
Effects of chronic administration with high doses of testosterone propionate on behavioral and physiological parameters in mice with differing basal …
2003
The effects of testosterone propionate, an anabolic-androgenic steroid, on the behavior displayed during a social encounter by gonadally intact male mice were investigated. Animals were distributed into three groups according to their attack latency in a pre-screening test (high-, moderate-, and low-attacking mice) and each group received weekly injections of 60 or 120 mg/kg of testosterone or sesame oil for 10 weeks. Behavioral tests were then carried out. Afterwards, organs were weighed and blood samples collected in order to obtain hormonal data. Treatment had a differential impact on attack in the three groups of animals. Only the high-attacking testosterone-treated mice showed lower to…
Retention-structure relationship studies for some steroidal hormones in micellar liquid chromatography
2000
The retention behavior in a C18 column of fourteen steroidal hormones (clostebol acetate, dehydrotestosterone, dydrogesterone, medroxyprogesterone, medroxyprogesterone acetate, methandienone, methyltestosterone, nandrolone, nandrolone decanoate, progesterone, testosterone, testosterone enanthate, testosterone propionate and, stanozolol) with pure micellar mobile phases prepared from sodium dodecyl sulfate (SDS), and hybrid mobile phases comprising SDS-acetonitrile and SDS-pentanol, was correlated with their octanol-water partition coefficients. Similar correlations were found with retention data obtained by other authors in gas chromatography, conventional reversed-phase liquid chromatograp…
Evaluation of anti-androgenic activity of di-(2-ethylhexyl)phthalate
2005
International audience; DEHP is a widely used platiciser in the manufacture of PVC-based materials. It is known to disrupt the reproductive tract development in male rats. We have performed the Hershberger assay with DEHP on an immature castrated rat model to check if DEHP antagonise the testosterone propionate androgenic effect on the accessory sex organs development. DEHP significantly decreased the BC/LA muscles, the prostate, and the seminal vesicles relative weights from 100, 200, and 400 mg/kg bw/day, respectively. DEHP increased the liver relative weight from 100 mg/kg bw/day. A study was also performed on MDA-MB453 cell line stably transfected with pMMTVneo-Luc with DEHP and its maj…
Resolution of mixtures of steroidal hormones with micellar eluents of sodium dodecyl sulphate and acetonitrile or pentanol
2000
An optimization strategy was applied to explore the capability of hybrid micellar eluents of sodium dodecyl sulphate (SDS), using acetonitrile or pentanol as modifiers, to resolve mixtures of eleven steroids showing a wide range of hydrophobicity (clostebol acetate, dehydrotestosterone, dydrogesterone, medroxyprogesterone, medroxyprogesterone acetate, methandienone, methyltestosterone, progesterone, testosterone, testosterone enanthate and testosterone propionate). The accurate prediction of the retention behaviour of the steroids, with relative errors in the 0.8–1.7% and 0.4–2.9% ranges for SDS-acetonitrile and SDS-pentanol eluents, respectively, demonstrated the reliability of the methodo…
Topical canrenoic acid. Quantification of the antiandrogenic activity in the hamster flank organ
1991
: The topical antiandrogenic activity of potassium canrenoate (CAK), compared with that of spironolactone (SP), was assayed in vivo in female golden Syrian hamsters whose flank organs were stimulated by subcutaneous administration of testosterone propionate. Sebaceous glands and hair were measured by a computerized image analyzer. Pigmented spots, sebaceous gland areas, and the diameter of hairs of the treated flank organs were smaller in the groups that received CAK (1·6 mg/day) and SP (0·4 mg/day). The authors' results showed that CAK may act as a topical antiandrogen on the hamster flank organ when applied in concentrations four times greater than the minimal active dosage of SP. Potassi…
Lack of effects of anabolic-androgenic steroids on locomotor activity in intact male mice.
1999
Anabolic-androgenic steroid abusers have reported hyperactivity euphoria, and decreased fatigue, among other behavioral effects. It has been suggested that the effects of these substances on the central nervous system are similar to those of psychostimulants; however, the influence of steroids on general locomotor activity in laboratory animals is not well understood, especially how noncastrated male rodents are affected. In this study, spontaneous locomotor activity displayed by gonadally intact male mice submitted to several experimental conditions was analyzed. Different housing conditions (individual or cohabiting with a female), diverse steroids (testosterone propionate, nandrolone de…
A research on the action of testosterone propionate and of ciproterone acetate on the mouse-killing behaviour of the adult rat.
1979
AbstractData demonstrate exactly that testosterone favour above all the killing behaviour of castrate males at birth in comparison with control males while ciproterone acetate turns out to be more effective in diminishing the killing behaviour of males having integral testicle and of androgenized females at birth in comparison with castrated males. The results point out that the killing behaviour of rats is affected by the presence of testosterone in the first days of life, and in the grown-up animal.
Effects of chronic treatment with testosterone propionate on aggression and hormonal levels in intact male mice.
1998
Effects of testosterone propionate, an anabolic-androgenic steroid (AAS), on aggression in gonadally intact male mice were examined. Animals were given weekly injections of 3.75, 7.5, 15, and 30 mg/kg of drug or sesame oil for 10 weeks. During the last 3 weeks, behavioral tests were conducted and at the end of the experiment, body, liver and testes weight and hormonal data were collected. The treatment had minimal behavioral and endocrine effects. It resulted in shorter latencies of 'threat' only in the last agonistic encounter, increases in testosterone levels and decreases in testes weight in a non-linear dose-dependant way. The action of treatment was different on threat and attack, the …
DETERMINATION OF STEROIDAL HORMONES IN URINE SAMPLES BY MICELLAR LIQUID CHROMATOGRAPHY FOLLOWING SOLID-PHASE EXTRACTION
2001
Steroidal hormones were determined in spiked urine samples using micellar mobile phases of sodium dodecyl sulphate (SDS)-pentanol, solid-phase extraction (SPE), and detection in the UV region. In the optimized procedure, a 10 mL aliquot of urine sample is loaded into a C18 cartridge and washed with 2 mL of 50:50 (v/v) methanol-water, followed by 200 μL of 70:30 (v/v) methanol-water. The retained steroids are eluted with 2 mL of methanol and the eluate evaporated to dryness under nitrogen at 50°C. The residue is redissolved with 200 μL of the micellar mobile phase used in the chromatographic separation and injected into the chromatograph. The performance of the procedure was checked for 13 s…
